Vimentin is a type III intermediate filament protein encoded by the VIM gene. It is predominantly expressed in mesenchymal cells such as fibroblasts, endothelial cells, macrophages, melanocytes, Schwann cells, and lymphocytes. As one of the major components of the cytoskeleton—alongside microtubules and actin filaments—vimentin provides structural support by maintaining cell shape and mechanical stability. Beyond its traditional role as a static scaffold within the cytoplasm, recent research has revealed that vimentin forms dynamic networks involved in regulating signal transduction pathways, organelle positioning/transportation within the cell, modulation of integrins affecting adhesion/migration properties during processes like wound healing or metastasis formation. It plays an essential part during epithelial-to-mesenchymal transition—a key event both in embryonic development and cancer metastasis—and can be found at the surface or secreted under certain conditions such as immune responses. Pathogens may exploit surface-expressed or secreted forms for infection strategies. Clinically it serves primarily as a diagnostic/prognostic biomarker rather than a direct therapeutic target; however experimental compounds like Withaferin A have been shown to disrupt its function with potential anti-tumor effects.
