Overview

Viral envelope-associated processes encompass the critical stages of the viral life cycle involving the lipid bilayer and its embedded glycoproteins. These processes include viral attachment to host receptors, membrane fusion, entry into the host cell, and the eventual budding of new virions from the host cell membrane (Nature Reviews Microbiology, 2004). Because these steps are essential for the propagation of enveloped viruses such as HIV, Influenza, and Coronaviruses, they represent major therapeutic targets (PubMed, PMID: 15175301). Drugs like enfuvirtide and maraviroc interfere with these processes by blocking the conformational changes required for fusion or by masking the binding sites on host cells (PubChem). Targeting these processes is a key strategy in preventing the initiation of infection and reducing viral spread within the host (NIH, 2023). However, the high mutation rate of viral envelope proteins often leads to the rapid emergence of resistant strains, posing a significant challenge for long-term treatment efficacy (DrugBank).
