Overview

The viral envelope lipid membrane is a host-derived lipid bilayer that surrounds the genetic material and capsid of many types of viruses, including HIV, influenza, and coronaviruses. This membrane is acquired during the budding process from host cell membranes (such as the plasma membrane, ER, or Golgi) and is embedded with viral glycoproteins essential for host cell recognition and entry. Because the envelope is critical for the structural integrity and infectivity of enveloped viruses, it serves as a significant therapeutic target. Drugs like docosanol work by interfering with the fusion process between the viral envelope and the host cell, while experimental broad-spectrum antivirals like LJ001 specifically target the biophysical properties of the viral lipid bilayer to prevent entry. However, a major challenge in targeting this structure is achieving selectivity to avoid damaging the host's own cellular membranes.

Mechanism of action

Drugs targeting the viral envelope typically act by disrupting the lipid bilayer integrity, inhibiting the fusion of the viral envelope with the host cell membrane, or inducing lipid oxidation to render the virus non-infectious.
