Virus replication proteins are viral enzymes or accessory factors encoded by the viral genome that participate in the duplication of viral genetic material within host cells[1][3]. They include RNA-dependent RNA polymerases (RdRp), DNA polymerases, reverse transcriptase, helicases, and accessory factors that form the replication complex[3]. These proteins are central to the viral life cycle and are often highly specialized, dependent on the unique replication strategies of different viruses (e.g., DNA, RNA, retroviruses)[1][2]. They are also prime therapeutic targets, although the name "virus replication protein" does not denote a unique protein; instead, drug discovery focuses on specific viral polymerases or replication complexes[4]. High mutation rates, especially in RNA viruses, present therapeutic challenges and drive drug resistance[2].
