Viral membrane fusion is a critical step in the entry of enveloped viruses into host cells, mediated by viral fusion proteins (VFPs). This process enables the viral genome to access the host cell's interior, initiating infection. VFPs undergo conformational changes triggered by receptor binding, pH changes, or proteolytic cleavage, leading to the insertion of a fusion peptide into the host cell membrane, membrane apposition, hemifusion, pore formation, and ultimately, release of the viral genome into the cytoplasm. Viral membrane fusion represents a universal step required for the life cycle of enveloped viruses and is an attractive target for antiviral drugs and immunotherapies.
