Viral mRNA guanylyltransferase is an enzyme that catalyzes the addition of a guanosine cap to the 5' end of viral mRNA transcripts. This capping process is essential for mRNA stability and translation efficiency. The enzyme transfers GMP from GTP to the 5'-triphosphate or diphosphate terminus of RNA, forming the cap structure G(5')ppp(5')N. Examples include NSP14 in SARS-CoV-2 and the guanylyltransferase of vaccinia virus. Inhibitors targeting this enzyme could disrupt viral mRNA capping, affecting viral replication and translation.
