Overview

Viral replication machinery and associated host factors refer to the collective set of viral-encoded proteins and host cellular components that facilitate the viral life cycle within a host cell (Nature Reviews Microbiology, 2021). The viral machinery typically includes essential enzymes such as RNA-dependent RNA polymerases (RdRp), proteases, and helicases, which are responsible for replicating the viral genome and processing viral polyproteins (NIH, 2023). Associated host factors are endogenous proteins, such as the ACE2 receptor for entry or ribosomal machinery for translation, that viruses hijack to replicate efficiently (PubMed, 2022). This broad category is a primary focus for antiviral drug development, where direct-acting antivirals (DAAs) target viral enzymes, while host-directed therapies aim to disrupt the cellular environment the virus depends on (Frontiers in Microbiology, 2020). Because this term encompasses a wide array of distinct molecular targets across various virus families, it is generally considered a functional group or pathway rather than a single therapeutic target.
