Overview

Virulence factor regulator (Vfr) is a global transcription factor in Pseudomonas aeruginosa that belongs to the cAMP receptor protein (CRP) family. It acts as a master regulator of bacterial pathogenesis by controlling the expression of over 200 genes, including those involved in the Type III secretion system, quorum sensing, and twitching motility. Vfr activity is primarily regulated by the intracellular concentration of cyclic AMP (cAMP), which binds to the protein and induces a conformational change that allows it to bind specific DNA sequences. Because Vfr is essential for the full virulence of P. aeruginosa in various infection models, it is considered a high-priority target for the development of anti-virulence therapies. Inhibiting Vfr offers a strategy to attenuate the pathogen's ability to cause tissue damage and evade the immune system without directly killing the bacteria, potentially reducing the selective pressure for antibiotic resistance.
