The vitamin K-dependent clotting pathway involves a group of coagulation factors (II, VII, IX, X) and regulatory proteins (Protein C, S, Z) that require vitamin K for gamma-carboxylation of glutamate residues. This modification is crucial for calcium binding and membrane interaction, essential steps in the coagulation cascade. Dysfunction in this pathway can lead to bleeding or thrombotic disorders. Warfarin inhibits this pathway via inhibition of VKORC1.
