The synthesis of vitamin K-dependent clotting factors is a critical biochemical process in hemostasis, relying on the presence of phytomenadione (vitamin K1). This pathway enables the liver to produce several coagulation proteins essential for normal blood clotting. The process involves gamma-carboxylation of specific glutamate residues in Factors II, VII, IX, X, and Proteins C and S, which is strictly dependent on vitamin K1. Warfarin inhibits this pathway by blocking vitamin K epoxide reductase.
