Vitamin K-dependent coagulation factors (II, VII, IX, and X) are essential serine proteases involved in the blood clotting process. They require vitamin K for post-translational γ-carboxylation, enabling calcium binding and interaction with phospholipid membranes. Deficiencies or inhibition, such as by warfarin, lead to impaired hemostasis and increased bleeding risk. Mutations can result in bleeding disorders or thrombosis.
Vitamin K antagonists inhibit γ-carboxylation of coagulation factors, reducing their ability to bind calcium and participate in the coagulation cascade.
