CACNA2D1 encodes the voltage-dependent calcium channel subunit alpha-2/delta-1, a key auxiliary component of the voltage-gated calcium channel complex. It regulates calcium current density and modulates activation/inactivation kinetics of the main pore-forming subunit. Dysregulation is linked to various diseases, and it is a molecular target for gabapentinoid drugs used to treat epilepsy and neuropathic pain.
Gabapentinoids bind to the α₂δ₁ and α₂δ₂ isoforms of the voltage-dependent calcium channel, modulating calcium influx and reducing neurotransmitter release. Ziconotide blocks N-type calcium channels.
