Overview

The voltage-gated calcium channel alpha(2)delta (α2δ) subunit is an auxiliary protein component of high-voltage-activated (HVA) calcium channels, essential for Ca²⁺ influx into excitable cells. It regulates channel trafficking, surface expression, and biophysical properties. Four isoforms (α2δ-1 to α2δ-4) exist, encoded by CACNA2D1-4. The α2δ subunit influences synaptic function, neurotransmitter release and is implicated in neurological disorders like neuropathic pain and epilepsy. Gabapentinoids target α2δ-1 and α2δ-2 for pain management.
