The voltage-gated calcium channel alpha-2-delta-1 subunit is an *extracellular auxiliary protein* critical for the function of high-voltage activated calcium channels. Gabapentinoid drugs bind directly to the alpha-2-delta-1 subunit, inhibiting its function, which reduces the trafficking and activity of voltage-gated calcium channels, ultimately suppressing neurotransmitter release and dampening synaptic excitability.
