The voltage-gated calcium channel alpha-2-delta (α2δ) subunit is an auxiliary protein component of high-voltage activated calcium channels. It plays a critical role in modulating the trafficking, density, and biophysical properties of these channels at the plasma membrane. It is encoded by four genes: CACNA2D1, CACNA2D2, CACNA2D3, and CACNA2D4. The α₂δ₋₁ and α₂δ₋₂ isoforms are molecular targets for gabapentinoid drugs like gabapentin and pregabalin, used for neuropathic pain and epilepsy. Recent research indicates that some alpha‐2/delta proteins interact directly with glutamate receptors or extracellular matrix proteins—suggesting broader roles beyond classic channel modulation.
