Voltage-gated calcium channel (VGCC) complexes are multi-subunit transmembrane proteins that mediate the influx of calcium ions into excitable cells in response to membrane depolarization [1, 5]. These complexes typically consist of a pore-forming alpha-1 subunit, which determines the channel's primary biophysical and pharmacological properties, and several auxiliary subunits including beta, alpha-2-delta, and sometimes gamma [2, 4]. The auxiliary subunits are critical for regulating the trafficking of the alpha-1 subunit to the plasma membrane and modulating its gating kinetics [1, 11]. VGCCs are classified into several subtypes (L, N, P/Q, R, and T-type) based on their voltage-activation thresholds and sensitivity to specific blockers [8, 12]. They play essential roles in physiological processes such as excitation-contraction coupling in muscle, neurotransmitter release at synapses, and the regulation of gene expression [12, 13]. Dysregulation of these channels is implicated in a variety of conditions, including hypertension, chronic pain, epilepsy, and Lambert-Eaton myasthenic syndrome [1, 12]. Consequently, they serve as major therapeutic targets for diverse drug classes, ranging from antihypertensive dihydropyridines to analgesic gabapentinoids [2, 6].
