The voltage-gated calcium channel protein subunit alpha1S (Cav1.1) is the pore-forming subunit of the L-type voltage-dependent calcium channel primarily located in skeletal muscle cells. It plays a crucial role in excitation-contraction coupling by sensing membrane depolarization and mechanically interacting with the ryanodine receptor (RyR1) on the sarcoplasmic reticulum, triggering calcium release and muscle contraction. Mutations in Cav1.1 are associated with hypokalemic periodic paralysis, thyrotoxic periodic paralysis, and malignant hyperthermia susceptibility. It is a target of dihydropyridine drugs.
