Von Willebrand factor A domain-containing protein 5A (VWA5A), also known as BCSC1 or LOH11CR2A, is a nuclear-localized tumor suppressor protein implicated primarily in the suppression of cancer cell proliferation, migration, and invasion[1][2][3][5][7]. Reduced expression or genetic loss of VWA5A is correlated with higher malignancy, enhanced metastatic potential, and poor survival, particularly in hormone receptor–positive and triple-negative breast cancers[1][3]. VWA5A is located in a chromosomal region (11q23–q24) frequently deleted in multiple cancer types, and may contribute to genomic stability in concert with nearby genes such as CHEK1[1]. In breast cancer, lower VWA5A levels are linked to advanced tumor stage, lymphatic invasion, and shorter time to metastasis, making VWA5A protein expression in tissues a prognostic biomarker for metastasis and survival outcomes. While identified primarily as a tumor suppressor, there are no approved drugs directly targeting or modulating VWA5A, and its main therapeutic utility is as a diagnostic/prognostic biomarker rather than as a classic druggable target[1][2][3][5][6][7].
