VPS10 domain-containing receptor SorCS1 (SORCS1) is a type I transmembrane protein of the VPS10 domain receptor family, predominantly expressed in the central nervous system but also implicated in metabolic tissues[2][3][5]. SORCS1 regulates intracellular trafficking of membrane proteins, particularly critical for synaptic function and integrity. Alternative splicing produces isoforms with distinct cytoplasmic tails, conferring varied trafficking and cell-surface expression[4][6]. SORCS1 interacts with proteins such as Rab11FIP5 (Rip11), SorL1, and Vps35, orchestrating endosomal sorting and recycling[1][6]. Beyond the brain, SORCS1 modulates metabolic pathways, with genetic variants linked to diabetes and systemic disease risk[2][6]. Its dysfunction or altered regulation is associated with Alzheimer’s disease (via effects on amyloid precursor protein), glycemic disorders, and some cancers—a basis for its consideration as a biomarker and potential therapeutic target[3][6][8].
