VPS10 domain-containing receptor SorCS1 (SORCS1) is a type I transmembrane protein of the VPS10 domain receptor family, predominantly expressed in the central nervous system but also implicated in metabolic tissues[2][3][5]. SORCS1 regulates intracellular trafficking of membrane proteins, particularly critical for synaptic function and integrity. It sorts neuronal receptors such as neurexin 1α, influencing the balance between axonal and dendritic localization, thereby affecting neuronal connectivity and signaling[1][6]. Alternative splicing produces isoforms with distinct cytoplasmic tails, conferring varied trafficking and cell-surface expression[4][6].
