VPS35 endosomal protein sorting factor like (VPS35L) is a protein-coding gene encoding a core component of the retriever complex, a heterotrimeric complex related to the retromer cargo-selective complex. VPS35L is essential for retromer-independent retrieval and recycling of multiple cargo proteins, especially those mediating cell migration, adhesion, nutrient supply, and cell signaling, by coupling with SNX17 and interacting with CCC and WASH complexes on endosomes. It plays a role in the maintenance and trafficking of cell-surface proteins, as well as copper-dependent trafficking of ATP7A between the trans-Golgi network and peripheral vesicles. Mutations in VPS35L cause Ritscher-Schinzel syndrome 3, a multisystem neurodevelopmental disorder, but current evidence does not support a role as a direct therapeutic target (such as a druggable receptor, enzyme, or transporter)[1][3][4].
