Overview

The VRC01-class B cell receptor refers to a set of B cell receptors (BCRs) whose immunoglobulin genes encode the precursors of VRC01-class broadly neutralizing antibodies (bnAbs) against HIV-1. These BCRs use a heavy chain variable region derived from the human immunoglobulin VH1-2*02 gene, and a light chain featuring a rare, short (typically 5 amino acids) CDRL3 region. VRC01-class B cell receptors are characterized by a unique mode of antigen recognition, mimicking the interaction of human CD4 with the HIV-1 envelope glycoprotein gp120 at the "CD4 binding site" (CD4bs). They are noteworthy for their potential to develop into potent bnAbs through affinity maturation, enabling the neutralization of a broad range of HIV-1 strains. VRC01-class B cell receptors themselves are not direct therapeutic targets but are critical in HIV vaccine research, as their successful activation and maturation by germline-targeting immunogens is a central strategy in efforts to elicit VRC01-like bnAbs via vaccination. Their study informs the design of sequential immunization regimens intended to shepherd rare B cell precursors through the somatic mutations required for broad and potent HIV-1 neutralization. Additional details: - VRC01-class B cell receptors are rare among naïve human B cells, due to the required genetic features being uncommon and further shaped by central B cell tolerance mechanisms. - Authentic VRC01-class B cell receptors undergo extensive somatic hypermutation (up to 40% from germline) to achieve neutralization breadth; affinity maturation is driven in germinal centers after antigen encounter and subsequent immunization. - VRC01-class B cells are not associated with disease except insofar as antibodies derived from them can neutralize HIV-1. - There are currently no drugs that directly target or modulate these BCRs; instead, specifically engineered immunogens (such as eOD-GT8) are used to selectively activate them as part of vaccine development strategies. - As a concept, the "VRC01-class B cell receptor" is a scientific tool, not a therapeutic target, and thus most drug discovery data fields do not apply. Strictly speaking, "VRC01-class B cell receptor" is not a standard molecular or drug target (such as a receptor, enzyme, ion channel, etc.), but rather a classification of antibody gene usage and functionally relevant B cell clones.
