Overview

VRC01-class precursor B cell receptors are a distinct subset of immunoglobulin receptors carried by naïve B cells capable of maturing into cells that produce VRC01-class broadly neutralizing antibodies against HIV-1. These receptors feature a heavy chain encoded by the IGHV1-2*02 allele paired with a light chain possessing an unusually short or glycine/serine-rich complementarity-determining region 3 (CDRL3). Their primary biological function is to recognize the CD4 binding site (CD4bs) of the HIV-1 envelope glycoprotein (Env). Engagement by germline-targeting immunogens such as eOD-GT8 leads to the activation and maturation of these naïve B cells, initiating the adaptive immune response necessary for the generation of potent and broadly acting HIV-1 neutralizing antibodies. VRC01-class precursor B cell receptors represent a key therapeutic target in the field of HIV vaccine development, with efforts focused on priming and activating these cells to enable long-term protective immunity.
