Walnut allergen proteins are a group of proteins found in English walnut (Juglans regia) and Black walnut (Juglans nigra) that are responsible for triggering IgE-mediated hypersensitivity reactions in sensitized individuals. The most clinically significant allergens include Jug r 1 (a 2S albumin), Jug r 2 (a 7S globulin), Jug r 3 (a lipid transfer protein), and Jug r 4 (an 11S globulin) (Source: WHO/IUIS Allergen Nomenclature). These proteins primarily function as seed storage proteins, providing essential nutrients during germination, or as defense proteins within the plant (Source: UniProt). In humans, these allergens are recognized by the immune system as foreign threats, leading to the production of allergen-specific IgE antibodies and subsequent mast cell degranulation upon re-exposure. Walnut allergy is one of the most common tree nut allergies and is frequently associated with severe, life-threatening anaphylaxis due to the high thermal and proteolytic stability of these proteins (Source: NIH/NIAID). Therapeutic approaches targeting these proteins focus on oral immunotherapy (OIT), where controlled, escalating doses of the allergen are administered to induce desensitization and increase the threshold for allergic reactions (Source: ClinicalTrials.gov, NCT03737227). Additionally, monoclonal antibodies like omalizumab are being investigated as adjunct therapies to neutralize IgE and improve the safety profile of walnut-specific interventions.
Allergen-specific immunotherapy (AIT) involves the controlled administration of increasing doses of walnut allergen proteins to induce immunological desensitization and long-term tolerance. This process shifts the immune profile from a Th2-biased IgE response to a Th1/Treg-mediated response, characterized by the production of protective IgG4 antibodies and reduced activation of mast cells and basophils (Source: PubMed, PMID: 30102904).
