WASH complex subunit 1 (WASHC1) is a core component of the WASH complex, a multi-protein assembly that regulates actin polymerization at the surface of endosomes as a nucleation-promoting factor, primarily through activation of the Arp2/3 complex[1][3][4]. WASHC1 functions in the regulation of endosomal protein sorting, trafficking, and maintenance of endolysosomal system architecture, supporting critical processes such as endocytic cargo recycling and lysosomal acidification[2][3]. Emerging evidence indicates that, beyond its cytoplasmic functions, WASHC1 also plays roles in the nucleus, including regulation of gene transcription, DNA repair, and safeguarding genomic stability under conditions of replication stress, partly by interacting with the MCM2-7 complex and assisting in the loading of DNA replication-origin components[1]. It is not a common drug target or therapeutic receptor, enzyme, transporter, or ion channel, but disruptions in WASHC1/complex function have been implicated in pathological processes such as cancer by promoting chromosomal instability[1][2][4][6].
