Overview

The term 'Watermelon' does not refer to a recognized canonical therapeutic target, receptor, or enzyme in human biology or clinical pharmacology. In scientific literature and biotechnology, it most commonly describes two distinct research tools: an experimental lineage-tracing system and a computational drug discovery algorithm. The 'Watermelon system' is a high-complexity lentiviral barcode library used to track the fate of individual cancer cells, specifically identifying 'persister' lineages that exhibit non-genetic drug resistance during treatments such as EGFR inhibition [1, 2, 5]. Additionally, 'Watermelon' is the name of a computational in silico fragment-based approach designed to develop receptor-based pharmacophore models for virtual screening of novel inhibitors [4, 7, 17]. While the plant species Citrullus lanatus (watermelon) contains bioactive compounds like L-citrulline and lycopene that are studied for nutritional and metabolic health, there is no human protein or molecule canonically named 'Watermelon' that serves as a drug target [9, 12].
