Overview

WD repeat- and FYVE domain-containing protein 4 (WDFY4) is a large cytoplasmic scaffold protein characterized by the presence of BEACH, PH, WD40, and FYVE domains. It is predominantly expressed in immune organs and is essential for antigen cross-presentation by CD8α+ classical dendritic cells, thereby regulating CD8+ T cell responses critical for antiviral and antitumor immunity. WDFY4 is also involved in B-cell development, non-canonical autophagy, and vesicular trafficking, functioning as a molecular scaffold for protein complexes associated with endosomes and the endoplasmic reticulum. Genetic variants in WDFY4 are linked to autoimmune disorders and have been implicated as prognostic markers in several cancers, although no drugs are currently known to target this protein directly.
