Overview

WD repeat and SOCS box-containing protein 1 (WSB1) is a substrate recognition protein within an E3 ubiquitin ligase complex. It contains multiple WD40 domains at the N-terminus and a SOCS box at the C-terminus. WSB1 is involved in targeted protein ubiquitination and degradation, affecting key proteins involved in cell cycle, hypoxia response, glucose metabolism, immune regulation, and neuroprotection. It is transcriptionally regulated in response to hypoxia via HIF-1 and has been implicated as a regulator of cancer cell proliferation and metastasis through degradation of tumor suppressor proteins (e.g., VHL, ATM). WSB1 also affects thyroid hormone activation through modulation of deiodinase D2. Dysregulation of WSB1 has been linked to poor prognosis in cancers and may modulate cellular responses relevant to neurodegeneration and metabolic disease.
