Overview

WD repeat-containing and planar cell polarity effector protein (WDPCP) is a cytoplasmic WD40 repeat protein that acts as a key effector in the planar cell polarity (PCP) signaling pathway and is essential for ciliogenesis[1][2][3][5][6]. It plays a central role in organizing the cytoskeleton—particularly regulating septin localization—and controls both cilia formation and collective cell movements[1][3][5]. WDPCP is also required for maintaining the stability of the epithelial cell cortex and plasma membrane via interactions with septins and actomyosin[3][5]. Mutations in WDPCP are implicated in Bardet-Biedl syndrome type 15, Meckel-Gruber syndrome, and are associated with ciliary dysfunction in other settings such as chronic rhinosinusitis[1][2][3][5][6]. Despite its crucial developmental and structural cellular roles, it is not currently considered a direct therapeutic target or drugged protein[1][2][6].
