Overview

WD repeat-containing protein 37 (WDR37) is part of the WD repeat protein family, characterized by minimally conserved ~40 amino acid repeats, typically facilitating multiprotein complex assembly. WDR37 contains seven WD40 motifs, predicted to form a structurally stable seven-bladed β-propeller, serving as a scaffold for protein interactions[1][3][5][6]. It interacts strongly with phosphofurin acidic cluster sorting proteins (PACS1 and PACS2), suggesting a coordinated role in neuro-ocular development[1][2]. Its physiological functions include regulation of cell cycle, signal transduction, apoptosis, gene expression, and ER calcium handling within lymphocytes[5]. Pathogenic variants in WDR37 result in a novel multisystemic syndrome affecting neurological, ocular, gastrointestinal, genitourinary, and cardiac development[1][2][5]. While WD repeat domain proteins are being explored as a novel target class in drug development, WDR37 itself is not yet a direct target for any therapeutic interventions[6]. Note: WDR37 is involved in protein scaffolding and complex assembly essential for diverse cellular processes but is not an established receptor, transporter, enzyme, or direct drug target in current biomedical literature[2][6].
