Overview

WD repeat-containing protein 46 (WDR46) is a nucleolar scaffold protein that is essential for the spatial organization of ribosomal RNA (rRNA) processing machinery within the nucleolus[1][3][5][6][8]. It is a core part of the small-subunit (SSU) processome, which is responsible for biogenesis of the small ribosomal subunit[3][9]. WDR46 interacts with key proteins such as nucleolin and DDX21, facilitating their correct compartmental localization within the nucleolus[1][5][6][8]. The protein is characterized by WD40 repeats, forming a β-propeller structure typical of this large protein family, which generally function as molecular scaffolds for complex assembly without enzymatic or receptor activity[2][4]. There is currently no evidence that WDR46 has enzymatic function or serves directly as a therapeutic target. WDR46 has been weakly associated with certain conditions such as aspirin intolerance with nasal polyps and asthma, but these roles are associative rather than therapeutic or causal[3].
