Overview

WD repeat-containing protein 48 (WDR48) is a scaffold protein characterized by multiple WD-repeat motifs forming a beta propeller structure. It primarily functions to regulate and activate deubiquitinase complexes, notably USP1, USP12, and USP46, by binding to these enzymes and facilitating their activity. WDR48 plays a central role in DNA repair through modulation of FANCD2 deubiquitination, and it promotes homologous recombination in conjunction with RAD51AP1. Through its regulatory actions, it is implicated in cancer biology (tumor suppression via PHLPP1 stabilization) and neurodegenerative disease (via genetic mutations). WDR48 is not known to be targeted directly by approved drugs, though it modulates pathways essential for genome stability and cell signaling.
