Overview

WD repeat-containing protein 54 belongs to the WD40 repeat domain family, which are abundant scaffolding proteins involved in mediating protein-protein interactions[2][5]. WDR54 is notably upregulated in colorectal cancer, where its elevated expression correlates with worse disease-specific survival. Knockdown of WDR54 in cancer models inhibits cell aggressiveness and tumor growth, and increases sensitivity to SHP2 inhibitors, likely through regulation of key cancer-associated signaling pathways (AKT/ERK)[1][3]. The gene is implicated in regulatory functions associated with growth factor signaling, receptor internalization, and MAPK pathway regulation. No evidence suggests WDR54 is misspelled or incorrectly identified, but its molecular and druggability characterization outside cancer remains limited. No drugs are currently approved to target WDR54 directly, and safety concerns for WDR54-specific targeting have not yet been characterized in clinical settings.
