Overview

WD repeat-containing protein 8 (WRAP73) is a highly conserved member of the WD repeat protein family, characterized by repeated ~40 amino acid motifs that facilitate the assembly of multiprotein complexes. WRAP73 constitutively localizes to the centrosome, enriched at the proximal end of the mother centriole, and interacts with several key centrosomal and centriolar satellite proteins including SSX2IP, PCM1, CEP135, CEP290, and PIBF1/CEP90. Its precise roles include organizing spindle and microtubule architecture during mitosis, regulating docking of ciliary vesicles for ciliogenesis, and removing inhibitory complexes from the mother centriole to initiate the elongation of the axoneme and cilia assembly. Knockout and genetic studies reveal WRAP73 is critical for spindle integrity, ciliogenesis, and robust centrosomal function, particularly in concert with CEP135. Mutations in WRAP73 can cause isolated microspherophakia, a rare autosomal recessive developmental lens disorder. WRAP73 is also implicated in various basic cellular processes such as cell cycle progression, apoptosis, and ossification, based primarily on mouse models
