WD repeat domain 12 (WDR12) is a member of the WD repeat protein family characterized by the presence of WD motifs that typically facilitate multi-protein complex assembly. WDR12 is a core component of the PeBoW complex (alongside Pes1 and Bop1), which is essential for the maturation of the large ribosomal subunit (60S) by processing precursor rRNA into mature 5.8S and 28S forms—a critical step for ribosome assembly and cell proliferation[1][2][5]. WDR12 is predominantly localized in the nucleolus and contains seven WD repeats plus a ubiquitin-like N-terminal domain involved in protein-protein interactions[2][6]. Depletion or dysfunction of WDR12 impairs cell proliferation, disrupts rRNA processing, and can induce cell cycle arrest. WDR12 is ubiquitously expressed and implicated in cardiovascular diseases, being a genetic risk locus for coronary artery disease and myocardial infarction, with increased expression observed in failing hearts. However, it is not a classical druggable target, and there are no current drugs that specifically inhibit or modulate its function in clinical practice[2][5][7].
