WD repeat domain 43 (WDR43) is a protein-coding gene and essential ribogenesis factor that enables RNA binding and is highly involved in both ribosomal small subunit biogenesis and transcriptional regulation[1][5][4]. It acts by participating in the nucleolar processing of pre-18S ribosomal RNA and is required for the positive regulation of rRNA processing and transcription by RNA polymerase I. WDR43 is a part of the small subunit (SSU) processome, a critical ribonucleoprotein complex for the early steps of ribosome assembly. Emerging evidence suggests WDR43 also binds promoter-associated and nascent RNAs, facilitating release of elongation factors and paused RNA polymerase II, thereby regulating gene expression and maintaining stem cell pluripotency[1][5]. In disease states, WDR43 has been connected to cancer prognosis and immune cell infiltration, and is linked to ribosomopathy-related conditions like Diamond–Blackfan anemia[5]. No approved drugs are currently known to directly target WDR43, nor is there established therapeutic targeting of this molecule as of the most recent data[5].
