Overview

WD repeat domain 74 is a member of the WD repeat domain–containing protein family, localizing primarily in the nucleolus and nucleoplasm. It is involved in early steps of pre-ribosomal RNA processing, especially the maturation of the 60S ribosomal subunit, and regulates assembly of multiprotein complexes. WDR74 also acts as a transcriptional coactivator in certain signaling pathways, such as TGF-β/Smad. Aberrant expression or mutation of WDR74 promotes tumorigenesis in multiple cancers by modulating cell proliferation, apoptosis, and chemoresistance through pathways such as Wnt/β-catenin and RPL5–MDM2–p53. Its expression is linked to poor prognosis and enhanced cancer cell metabolic activity. Although no approved drugs currently target WDR74, its structure and role in cellular networks make it a promising candidate for therapeutic intervention[1][2][3][4].
