Overview

WD repeat domain-containing protein 83 (WDR83) is a member of the WD-40 protein family functioning as a molecular scaffold for a variety of multimeric protein complexes[1][2][7]. WDR83 is involved in organizing the extracellular signal-regulated kinase (ERK) signaling pathway, acting as a module to mediate and fine-tune ERK responses to different upstream signals. WDR83 also interacts with EGLN3/PHD3, modulating expression levels of hypoxia-inducible factor 1 (HIF-1) and linking hypoxic regulation with cell signaling and apoptosis. Additionally, WDR83 is implicated in RNA splicing and the formation of tight junctions by coordinating cellular polarity. Recent evidence suggests that WDR83 acts as a negative regulator of mTORC1 signaling in nutrient-rich conditions, thereby modulating cellular autophagy[2][7]. WDR83 is not a classic receptor, enzyme, or transporter, but rather a scaffold/adaptor protein involved in signal transduction and the organization of protein complexes, without current evidence supporting direct clinical drug targeting or use as a therapeutic biomarker[1][2][7].
