Overview

WDFY3 antisense RNA 1 (WDFY3-AS1) is a long non-coding RNA (lncRNA) transcribed in the antisense orientation relative to the protein-coding WDFY3 gene, located at chromosome 4q21.23[2]. WDFY3-AS1 does not encode a protein but acts as a regulatory RNA molecule involved in diverse cellular processes. In glioma, WDFY3-AS1 is significantly downregulated in high-grade tumors and its low expression is correlated with adverse prognosis, including poorer overall survival[2]. Functional and pathway analyses suggest this lncRNA is engaged in the regulation of synaptic transmission, glutamate receptor signaling, TNF signaling, cAMP pathway, and cell cycle processes[2]. WDFY3-AS1 may modulate gene expression by interacting with miRNAs and potentially regulates its sense gene WDFY3, which encodes a master conductor of autophagy and brain development[2][5]. It is investigated chiefly as a biomarker for glioma grade and prognosis; no direct drug interactions or established roles as a conventional therapeutic target (such as a receptor, enzyme, or transporter) have been reported[2].
