Serine/threonine-protein kinase WEE1 is a nuclear kinase that inhibits CDK1/cyclin B, preventing premature entry into mitosis. It plays a crucial role in the G2/M checkpoint, DNA damage response, and cell size control. WEE1 is a therapeutic target in cancer, with inhibitors designed to induce mitotic catastrophe in tumor cells.
Inhibition of WEE1 prevents inhibitory phosphorylation of CDK1, leading to premature mitotic entry and mitotic catastrophe, particularly in cancer cells with defective G1 checkpoints. Selectivity against tumor cells due to reliance on G2/M checkpoint.
