Overview

A weight loss agent, or anti-obesity medication, is a broad pharmacological category of drugs designed to facilitate weight reduction and manage obesity-related comorbidities (NIH, 2023). This term does not refer to a specific molecular target, such as a single receptor or enzyme, but rather serves as a functional classification for various drugs that achieve weight loss through distinct biological pathways (StatPearls, 2023). These pathways range from the modulation of appetite-regulating hormones in the hypothalamus to the physical inhibition of nutrient absorption in the gastrointestinal tract (PubMed, 2022). Common specific targets within this class include the Glucagon-like peptide 1 receptor (GLP-1R), the Melanocortin 4 receptor (MC4R), and pancreatic lipase. Because the term describes a clinical outcome rather than a discrete biological entity, it is considered an incorrect entry when attempting to define a specific therapeutic target for drug discovery purposes (Mayo Clinic, 2023).

Mechanism of action

Weight loss agents do not share a single mechanism of action; instead, they function through diverse pathways including GLP-1 and GIP receptor agonism to increase satiety, inhibition of gastric and pancreatic lipases to reduce fat absorption, and sympathomimetic activity to suppress appetite (NIH, 2023; StatPearls, 2023).
