Overview

Wilms tumor protein 1-interacting protein (WTIP) is an adapter or scaffold protein primarily involved in the regulation of gene transcription, acting as a corepressor for several transcription factors, and modulating cell cycle, cell adhesion, cytoskeleton organization, and cellular differentiation. It regulates miRNA-mediated gene silencing, negatively impacts the Hippo signaling pathway, and acts as a hypoxic regulator by facilitating the degradation of HIF1A. WTIP is expressed in various tissues including kidney, heart, and neural cells, and plays essential roles in development and physiologic homeostasis. In pathological contexts, notably non-small-cell lung cancer, it acts as a tumor suppressor by impairing AKT activation, promoting FOXO1 activity, and inducing cell cycle arrest through upregulation of cyclin-dependent kinase inhibitors and suppression of cyclin D1. Loss or downregulation of WTIP is associated with poor patient outcomes in cancer, and with diseases linked to podocyte dysfunction, developmental anomalies, and chromosomal syndromes.
