Overview

The Wnt signaling pathway is a highly conserved group of signal transduction pathways that play essential roles in embryonic development, cell fate determination, proliferation, migration, polarity, organogenesis, and adult tissue homeostasis. The pathway is initiated by Wnt ligands binding to Frizzled receptors and co-receptors like LRP5/6, leading to activation of downstream signaling cascades. These cascades include the canonical (Wnt/β-catenin) pathway, which regulates gene transcription, as well as non-canonical pathways involved in planar cell polarity and calcium signaling. Aberrant activation of the Wnt pathway is implicated in various diseases, including cancer.
