Wiskott-Aldrich syndrome protein family member 1 (WASF1), also known as WAVE1, is an actin-binding protein and key effector in the WAVE regulatory complex, mediating signals from tyrosine kinase receptors and Rho-family GTPases to the actin cytoskeleton[1][2][5]. WASF1 is essential for actin filament nucleation and reorganization through interactions with the Arp2/3 complex, crucial for cellular processes such as membrane ruffling, lamellipodia formation, and cell movement. It further participates in endocytic trafficking, mitochondrial dynamics, and cellular pathways influencing cell survival and chemoresistance. Loss-of-function mutations in WASF1 are associated with neurodevelopmental disorders characterized by intellectual disability, language impairment, and seizures[2][3].
