Overview

Wnt-3a is a secreted glycoprotein that activates the canonical Wnt/β-catenin signaling pathway by binding to Frizzled family receptors and the LRP5/6 co-receptor on cell surfaces[1][5][7]. This pathway is evolutionarily conserved and regulates cell proliferation, fate specification, stem cell maintenance, and embryonic development, with prominent roles in the formation of neural, skeletal, and hematopoietic tissues[1][3][5][7]. Abnormal Wnt3a pathway activation is implicated in cancer and aberrant tissue growth, making components of the pathway attractive targets for drug development, although precise targeting is challenged by its broad physiological roles and safety concerns[1][2][4][6][7].
