Overview

Wnt family members are a group of 19 secreted, cysteine-rich glycoproteins that act as essential ligands in the Wnt signaling pathway, which is highly conserved across species and vital for coordinating cell fate, proliferation, and tissue architecture during development and adulthood (PMID: 22354868, 30372336). These ligands bind to Frizzled (FZD) receptors and LRP5/6 co-receptors to trigger the canonical beta-catenin-dependent pathway or various non-canonical pathways, such as the Planar Cell Polarity (PCP) and Wnt/Ca2+ pathways (PMID: 28552352). Dysregulation of Wnt family signaling is a major driver of oncogenesis, particularly in colorectal, breast, and liver cancers, and also contributes significantly to fibrotic diseases and bone density disorders (PMID: 25964303, 31105436). Therapeutic strategies targeting these ligands include PORCN inhibitors, which prevent the post-translational palmitoylation required for Wnt secretion, and decoy receptors that sequester extracellular Wnts (PMID: 32430489). However, drug development is complicated by the pathway's critical role in normal stem cell niches, leading to significant safety concerns such as bone loss and intestinal toxicity (PMID: 28552352, 34536299).
