Overview

Wnt family member 4 (WNT4) is a cysteine-rich, secreted lipoglycoprotein of 351 amino acids that serves as a key ligand in several Wnt signaling pathways. WNT4 is essential for embryonic organogenesis, sexual differentiation, and tissue homeostasis, mediating both canonical (β-catenin dependent) and non-canonical (β-catenin independent) signal transduction by binding Frizzled family receptors. Its functions are highly context-dependent, modulating cell proliferation, differentiation, migration, and survival. WNT4 dysregulation is implicated in diverse disease states including cancer, bone and metabolic disorders, reproductive syndromes, and fibrosis. Although no drugs directly target WNT4 clinically, inhibition of its processing, secretion, or downstream signaling pathways represents an active area of research. Caution is advised in modulating WNT4 pharmacologically due to its wide-ranging roles in healthy tissue function.

Mechanism of action

Drugs may target WNT4 by preventing WNT4 ligand secretion or palmitoylation, inhibiting WNT4-induced activation of Frizzled receptors, or blocking downstream signaling (β-catenin, JNK, Ca2+-PKC, etc.). No approved drugs with defined mechanism against WNT4 itself.
