Wnt family member 4 (WNT4) (WNT4)

The Wnt family member 4 (WNT4) promoter is a regulatory DNA sequence that controls the expression of the WNT4 protein, a secreted signaling ligand essential for urogenital development and tissue homeostasis (UniProt: P56705; NCBI Gene: 54361). While the promoter itself is a site of intense research—particularly regarding its regulation by steroid hormones in endometriosis—the functional therapeutic target is typically the WNT4 protein or the broader Wnt signaling pathway (PubMed: 25673561). WNT4 acts as a ligand that triggers intracellular signaling cascades, such as the Wnt/beta-catenin pathway, which are often dysregulated in conditions like kidney fibrosis and ovarian cancer (PubMed: 28213345). Currently, pharmacological intervention primarily involves the use of Porcupine inhibitors, such as LGK974, which prevent the post-translational modification and secretion of WNT4 and other Wnt family members (ClinicalTrials.gov: NCT01351168). Therapeutic challenges in targeting this pathway include significant safety concerns related to bone density loss and gastrointestinal toxicity, given the critical role of Wnt signaling in maintaining adult stem cell niches (PubMed: 23681130).
