Wnt family member 8B (WNT8B)

Wnt family member 8B (WNT8B) is a secreted glycoprotein and a member of the Wnt protein family, which are crucial mediators of cell signaling through the canonical Wnt/β-catenin pathway[1][7]. WNT8B acts as a ligand for Frizzled family receptors and is fundamentally involved in embryonic development, patterning, regulation of cell fate, and organogenesis. It is expressed primarily in the developing brain and has tightly regulated expression patterns, frequently upregulated in several types of cancer, including liver, breast, pancreatic, and colorectal malignancies[1][5][7]. WNT8B’s function also encompasses suppression of early eye and retinal progenitor cell formation and plays roles in diseases such as epilepsy and certain congenital syndromes[1][2][3][7]. While no approved drugs specifically target WNT8B, its central role in activating canonical Wnt signaling makes it a potential therapeutic target and a prognostic biomarker, especially in cancer[5]. Modulating WNT8B or its pathway poses safety challenges due to the broad physiological functions of Wnt ligands[6].
