Overview

Wound protease activity refers to the collective function of multiple proteases, especially matrix metalloproteinases and serine proteases, which degrade and remodel the extracellular matrix in response to tissue injury. These enzymes are essential for normal wound healing, involved in removing damaged tissue, facilitating cell migration, and promoting new tissue formation. In chronic wounds, persistent inflammation, microbial contamination, and tissue debris drive excessive and prolonged protease activity, which impairs healing by destroying newly formed matrix and growth factors, thus perpetuating the wound's non-healing state. Therapeutic interventions often seek to restore the balance of protease activity using inhibitors, dressings, or anti-inflammatories, and elevated wound protease levels serve as useful biomarkers for delayed healing and may guide patient management.

Mechanism of action

Protease inhibition: Inhibiting or sequestering excessive protease activity to prevent extracellular matrix breakdown and promote wound healing. Anti-inflammatory action: Reducing inflammatory cytokine production and neutrophil infiltration lessens protease activation. Removal of damaging bacteria: Reducing bacterial burden can lower exogenous protease contributions.
