Overview

WW and C2 domain containing 2 (WWC2) is a human protein that functions as a regulator of the Hippo signaling pathway, also known as the Salvador-Warts-Hippo (SWH) pathway[2][4]. WWC2 contains two N-terminal WW domains (which mediate binding to proteins with L/PPxY motifs), an internal C2 domain for membrane association, and several C-terminal motifs for protein-protein interactions[2]. WWC2 is able to form homo- and heterodimers and primarily acts as a scaffold protein, coordinating the phosphorylation and suppression of YAP1, a major effector of the Hippo pathway, thus negatively regulating cell proliferation and organ growth[2][4]. Dysfunction or altered expression of WWC2 has been associated with cancer, particularly as a tumor suppressor through regulation of proliferation signals, and diseases such as Long QT syndrome[2][7]. There are currently no known drugs that directly target WWC2.
